PT-141: the brain’s role in desire
From melanocortin research to the 2019 FDA approval of Vyleesi: follow PT-141 (bremelanotide) through early studies, two phase three trials and the safety limits of its approved use.
English narration with optional English captions.

Sexual desire begins with signals in the brain. PT-141, also called bremelanotide, grew from research into a family of messengers called melanocortins.
By 2002, Palatin Technologies in Cranbury, New Jersey, was reporting human trials. A 2003 paper by Peter Molinoff, Annette Shadiack and colleagues described its development. That gives PT-141 more than two decades of clinical research history.
Bremelanotide is a ring-shaped peptide containing seven amino acids. It activates several melanocortin receptors, including receptors found in the brain. Its precise mechanism for improving low sexual desire remains unknown.
Early studies investigated sexual responses in men. Later development focused on women experiencing persistently low desire that caused distress.
Two phase three RECONNECT trials randomized 1,267 premenopausal women to bremelanotide or placebo for twenty-four weeks.
Women receiving bremelanotide reported greater improvements in desire and reductions in related distress. The trials did not show a significant advantage in the number of satisfying sexual events.
In June 2019, the FDA approved Vyleesi for acquired, generalized hypoactive sexual desire disorder in premenopausal women. By 2026, that represents seven years since U.S. approval. This is a specific indication, not a general sexual-performance claim.
Nausea, flushing and headache are important side effects. Blood pressure can temporarily rise, and the medicine is contraindicated in uncontrolled hypertension or known cardiovascular disease.
PT-141 connects decades of peptide research with a distinct approach to sexual health: studying desire, its biological signals, and the distress that can accompany its loss.
